Variations in protein binding of drugs in plasma and serum.

The free fractions of five drugs were determined in plasma treated with EDTA and in serum. The free fractions of phenytoin, sodium valproate, and phenobarbital in serum were significantly higher than in plasma (P less than 0.05). In contrast, the free fractions of carbamazepine and theophylline were significantly lower in serum than in plasma. Although we observed minor differences in the prote...

Predicting Protein Binding of Drugs Using Abraham Parameters: Effect of Ionization

Background and purpose: Protein binding (PB) is an important pharmacokinetic parameter in drug discovery and development. In past years Abraham parameters were used to predict some physicochemical and pharmacokinetic properties of drugs. But in these cases, the ionization of drugs in blood pH (7.4) was ignored. Recently, Abraham parameters of chemical compounds in ionized form are proposed. Als...

Protein binding of drugs.

Basic considerations involved in the interaction of drugs with proteins are reviewed. The important role of the concentration of unbound drug in determining therapeutic and toxicologic effects is emphasized. Ultra-filtration is suggested to be the method of choice for the determination of concentrations of unbound drug in plasma in clinical practice.

Designing, Optimization and Construction of Myelin Basic Protein Coding Sequence Binding to the Immunogenic Subunit of Cholera Toxin

Abstract Background and Objectives: Multiple sclerosis (MS) is a chronic inflammatory autoimmune disease. Mucosal feeding of myelin basic protein binding to the cholera toxin B subunit can reduce the intensity of the immune response in MS patients. Expression system, the domain composition of the fusion protein, accessibility of two domains, codon adaptation index (CAI) and GC contents are v...
